NORAD is a conserved and highly expressed lncRNA that regulates genomic stability, and it has been implicated in cancer and other disease processes.
Previous characterizations of NORAD were only based on sequence-based computational inferences and have identified a modular organization of NORAD composed of several repeat units (NRUs).
Here, we experimentally determined for the first time the secondary structure of NORAD using in-vitro enzymatic probing coupled to Illumina sequencing (nextPARS approach) to provide a thorough characterization of NORAD structure.
Our results suggest that the spacer regions provide structural stability to NRUs. Additionally, we uncover two previously unreported NRUs and determine the core structural motifs conserved across NRUs.
Given the current interest in understanding the structure, function and evolution of long, non-coding RNAs and the clinical relevance of NORAD, we think that eventually, this may open the door to design novel therapeutic strategies in the fight against cancer.
